Analysis on genetic polymorphism of avian leukosis virus subgroup J receptor gene NHE1

Abstract
To explore the potential genetic resistance loci in avian leukemia virus subgroup J( ALV-J) receptor gene NHE1,the sequence of NHE1 receptor gene was amplified by PCR technique,and the genetic polymorphism of NHE1 gene was analyzed. The results indicated that 16 SNP mutations,including c.309 G>C,c.11653 C>T,c.11689 G>A,c.11917 C>T,c.11946 C>T,c.13405 C>A,c.13429 T>C,c.13438 T>C,c.14108 A>C,c.14516 A>G,c.14522 G>A,c.14654 C>T,c.15834 G>A,c.15873 G>A,c.17560 C>G and c.17770 G>A,were found within the exon region of NHE1 gene in yellow broilers,these SNPs were synonymous with cSNP and the changes of amino acid were not caused by further analysis. In addition,SNPs and insertion/deletion mutations were also found within the intron region of NHE1 gene in yellow broilers,the AGGGCTGC was inserted between c.12239 and 12240 and the TGAAGCC was inserted between c.13114 and 13115 within the intron 2 of NHE1 gene,respectively,and the TGTCCTGTGTCCTCCCTGC among c.17073-17091 was deleted in intron 11 of NHE1 gene. Furthermore,6 SNPs c.309 G>C,c.11653 C>T,c.13405 C>A,c.13438 T>C,c.14108 A>C and c.14516 A>G within the exon region, as well as insertion/deletion mutations within the intron region that were same with yellow broilers,were found in NHE1 gene of the negative and positive chickens after inoculated by ALV-J. The results demonstrated that the genetic mutation loci found in receptor gene NHE1 may not be used as potential genetic resistance loci of ALV-J.
